应用系列稀释法PCR技术检测含5%携带TCRy基因重排的CCRF-CEM株细胞的模拟缓解期白血病骨髓的微量残留白血病细胞,敏感性为10 ̄_(-5)~10 ̄(-6)水平。经VPL加ADM、Vp16、VCR联合体外净化1小时,结果显示有3个对数级的杀伤作用。用体外克隆形成培养法观察该联合方案对急性期白血病患者白血病祖细胞集落(CFU-L)及正常骨髓造血干细胞集落(CFU-GM)的抑制作用,CFU-GM存活率为28.02%,而CFU-L为3.95%(P<0.01),提示有显著的选择性净化作用。
Serial dilution assay PCR technique was used to detect trace residual leukemia cells in leukemia bone marrow with 5% of CCRF-CEM strain cells carrying TCRy gene rearrangement. The sensitivity was 10 ~ (-5) ~ 10 ~ (-6). Level. The VPL plus ADM, Vp16, and VCR combination was purified in vitro for 1 hour and the results showed 3 logarithmic killing effects. The inhibitory effect of this combination regimen on acute leukemia patient leukemia progenitor cell colony (CFU-L) and normal bone marrow hematopoietic stem cell colony (CFU-GM) was observed by in vitro colony formation culture method. The survival rate of CFU-GM was 28.02%. The CFU-L was 3.95% (P<0.01), suggesting significant selective purification.
