为建立‘琼引1号’菠萝蜜快速增殖的有效方法,本研究以‘琼引1号’菠萝蜜长约0.50~1.00 cm单芽为外植体材料,结合前期调整培养基大量元素用量以及植物生长调节剂浓度等工作,通过逐步优化法,探讨了NH4NO3、Ca(NO3)2、6-BA+NAA不同配比浓度对菠萝蜜增殖效果的影响和PVP(polyvinylpyrrolidone)抑制外植体褐化的效果。结果表明,适宜‘琼引1号’菠萝蜜增殖培养和生长的NH4NO3浓度为412.50 mg/L,Ca(NO3)2浓度为531.00 m/L,6-BA浓度为1.50 mg/L,NAA浓度为0 mg/L,此时的菠萝蜜长势和增殖效果表现最佳,增殖系数达到3.17。研究还发现,PVP浓度达到0.30 g/L时可有效抑制褐化发生。菠萝蜜组培快繁技术的研发将有效加快菠萝蜜新品种的规模化种植及推广,同时还可为下一步的菠萝蜜分子育种提供技术基础。
