结、直肠癌是我国常见的恶性肿瘤之一,近年来有日趋增多之势。建立结、直肠癌细胞系,对于研究人类大肠癌细胞的生长、发展、转移和分化、逆转的规律及调节机制,提供良好的物质基础。大肠癌细胞培养难度较大,不易成功。我们采取将病人肿瘤组织先接种于裸鼠皮下,建立裸鼠移植瘤,再将移植瘤体外培养,在国内首次成功建立了裸鼠体内移植瘤及体外培养的细胞系,命名为THC-8901,现简介于下。癌组织来源于升结肠癌患者手术切除标本,病理组织学诊断为结肠粘液腺癌I级,接种于裸鼠皮下,至今已传13代,生长稳定并已冻存,病理检查各代肿瘤细胞组织形态,均符合人结肠高分化粘液腺癌。培养癌细胞取自结肠癌裸鼠移植瘤第4代瘤
Colorectal cancer is one of the most common malignancies in China and has been increasing in recent years. The establishment of colon and rectal cancer cell lines provides a good material basis for studying the growth, development, metastasis and differentiation and reversal of human colorectal cancer cells, and their regulatory mechanisms. Colorectal cancer cells are difficult to cultivate and are not easy to succeed. We took the patient’s tumor tissue first inoculated subcutaneously in nude mice, established a nude mouse transplanted tumor, and then in vitro cultured the transplanted tumor. The cancer tissue was derived from surgical specimens of patients with ascending colon cancer. Histopathological diagnosis was colonic mucinous adenocarcinoma grade I and inoculated subcutaneously in nude mice. It has been passed for 13 generations, has stable growth and has been cryopreserved, pathological examinations of various generations of tumor cell tissues.
