目的:观察钩藤提取液对吗啡诱导的大鼠条件性位置偏爱效应的影响。方法:SD雄性大鼠,取经过基线测定合格的40只大鼠,随机分为空白对照组、吗啡模型组(15 mg.kg-1,sc)、钩藤提取液低剂量(3 g.kg-1,ig)+吗啡组、钩藤提取液高剂量(9 g.kg-1,ig)+吗啡组,每组10只动物,给药5 d。采用倾向性程序训练大鼠,建立位置偏爱模型,观察钩藤提取液对大鼠在伴药箱(白箱)逗留时间的影响。结果:吗啡模型组训练5 d后,大鼠在白箱的逗留时间明显延长(与空白对照组比较,P<0.01),说明经过5 d训练,大鼠对吗啡形成了位置偏爱。给予钩藤提取液高剂量组(9 g.kg-1)能显著缩短大鼠在白箱的逗留时间(与模型组比较,P<0.05)。结论:钩藤提取液能够抑制吗啡诱导的条件性位置偏爱效应的形成,具有一定的抗药物精神依赖作用。
Objective: To observe the effect of Uncaria rhynchophylla extract on morphine-induced conditioned place preference in rats. Methods: SD male rats were randomly divided into blank control group, morphine model group (15 mg.kg-1, sc), Uncaria extract low dose (3 g.kg -1, ig) + morphine group, and Uncaria extract (9 g.kg-1, ig) + morphine group, with 10 animals in each group for 5 days. The predilection procedure was used to train the rats, and the position preference model was established to observe the effect of Uncaria rhynchophylla extract on the rat’s stay time in the companion box (white box). Results: After 5 days of training, morphine model rats in the white box stay significantly longer (compared with the blank control group, P <0.01), indicating that after 5 days of training, rats formed a preference for morphine. The high dose group (9 g.kg-1) treated with Uncaria rhynchophylla extract significantly shortened the stay time in white box (P <0.05 compared with the model group). Conclusion: Uncaria extract can inhibit morphine-induced conditioned place preference effect, and has a certain anti-drug-dependent effect.
