目的:探讨鲜鱼腥草挥发油对哮喘豚鼠肺组织干细胞因子(SCF)和KL-6表达的影响,分析鲜鱼腥草挥发油在减轻气道炎症可能的靶点及作用机制,为鲜鱼腥草挥发油制剂的开发提供实验依据。方法:对照组豚鼠给予生理盐水灌胃。将哮喘模型豚鼠分为模型组、高剂量组、中剂量组、低剂量组四组,模型组给予生理盐水灌胃,另外三组给予不同剂量鲜鱼腥草挥发油灌胃。采用酶联免疫吸附试验(ELISA)观察鲜鱼腥草挥发油对致敏攻击豚鼠肺组织中SCF、KL-6含量的影响。结果:哮喘模型组肺组织SCF含量升高(P<0.05),与高剂量组、中剂量组、低剂量组肺组比较差异有统计学意义(P<0.05)。哮喘模型组豚鼠肺组织KL-6含量增加(P<0.05),经鲜鱼腥草挥发油干预以后,高剂量组肺组织中KL-6的含量明显下降(P<0.01),中剂量组、低剂量组与模型组比较差异无统计学意义(P>0.05)。结论:哮喘豚鼠肺组织内SCF和KL-6表达增加,与哮喘发病密切相关。鲜鱼腥草挥发油能作用于哮喘豚鼠肺组织,能够降低哮喘豚鼠肺组织SCF和KL-6的表达,具有一定的减轻哮喘气道炎性反应的作用。
Objective: To investigate the effect of fresh Houttuynia volatile oil on the expression of stem cell factor (SCF) and KL-6 in lung tissue of asthmatic guinea pigs, and to analyze the potential target and mechanism of the volatile oil from Houttuynia cordata Thunb. In relieving airway inflammation. Volatile oil formulations to provide experimental basis for the development. Methods: The guinea pigs in the control group were given normal saline. The asthmatic model guinea pigs were divided into four groups: model group, high dose group, medium dose group and low dose group. The model group was given normal saline. The other three groups were given different doses of fresh Houttuynia cordata volatile oil. The enzyme-linked immunosorbent assay (ELISA) was used to observe the effect of fresh Houttuynia volatile oil on the content of SCF and KL-6 in the lungs of sensitized guinea pigs. Results: Compared with the high dose group, the middle dose group and the low dose group, the SCF content in the asthmatic model group was significantly increased (P <0.05). The content of KL-6 in the lung tissue of asthmatic model group was increased (P <0.05). The KL-6 content in lung tissue of high dose group was significantly decreased after the intervention of fresh Houttuynia cordata volatile oil (P <0.01) There was no significant difference between the dose group and the model group (P> 0.05). Conclusion: The expression of SCF and KL-6 in lung tissue of asthmatic guinea pigs is increased, which is closely related to the pathogenesis of asthma. Fresh Houttuynia volatile oil can act on the lungs of asthmatic guinea pigs, can reduce the expression of SCF and KL-6 in the lungs of asthmatic guinea pigs, and have a certain role in alleviating the airway inflammatory reaction of asthma.
