目的 建立一个小鼠胰腺外分泌实验系统 ,用以研究促分泌物质缩胆囊素 (CCK)和卡巴胆碱 (CCH)对小鼠胰腺外分泌的影响 ,并使用细胞信号酶系统的蛋白激酶C(PKC)拮抗剂Chelerythrine以了解CCK及CCH的可能作用机制。方法 前瞻性对照性研究。结果 CCK和CCH均能刺激小鼠胰腺腺泡使其分泌淀粉酶比对照组增加约 1倍 (7 0 2 %比 14 2 9% ) ,但CCK或CCH加Chelerythrine则使淀粉酶的分泌增加大约 2倍 (19 0 2 % )。结论 CCK及CCH的刺激胰腺外分泌作用涉及到PKC信号系统 ,且PKC拮抗剂Chelerythrine有增强CCK及CCH的刺激作用。
Objective To establish a mouse pancreatic exocrine experimental system to study the effects of secreted cholecystokinin (CCK) and carbachol (CCH) on the pancreatic exocrine of mice and use protein kinase C (PKC) Antagonist Chelerythrine to understand the possible mechanism of CCK and CCH. Methods Prospective controlled study. Results Both CCK and CCH stimulated the pancreatic acinar cells in mice to increase the amylase secretion by about 1-fold (70.02% vs 142.9%) compared with the control group, but CCK or CCH plus Chelerythrine increased the amylase secretion by about 2 times (19 0 2%). Conclusions CCK and CCH stimulate pancreatic exocrine function involves PKC signaling system, and PKC antagonist Chelerythrine can enhance the stimulation of CCK and CCH.
