目的:观察丹参素对大鼠肝细胞硫代乙酰胺(TAA)损伤的影响。方法:以体外原代培养大鼠肝细胞为模型,通过检测培养上清液中LDH逸出量和四甲基偶氮唑盐(MTT)显色法了解肝细胞的存活及增殖情况,同时检测细胞内、外SOD含量。结果:丹参素在一定范围内呈剂量依赖性地刺激肝细胞增殖,而且能阻止TAA引起的体外培养大鼠肝细胞LDH逸出的增加、增生指数的下降以及细胞内、外SOD活性的降低。丹参素对TAA引起的肝细胞损伤的保护作用高于市场销售的乳猪促肝细胞生长素(PHGF)。结论:丹参素具有抗TAA肝损伤和刺激损伤后肝细胞再生的作用
Objective: To observe the effect of Danshensu on rat hepatocyte thioacetamide (TAA) injury. Methods: Primary cultured rat hepatocytes were used as a model to detect the survival and proliferation of hepatocytes by detecting LDH release in culture supernatant and MTT colorimetric assay. Meanwhile, Intracellular and extracellular SOD content. Results: Danshensu stimulated hepatocyte proliferation in a dose-dependent manner in a certain range, but also inhibited the increase of LDH release, the decrease of proliferation index and the decrease of intracellular and extracellular SOD activity induced by TAA in rat hepatocytes cultured in vitro. Danshensu is more protective against TAA-induced hepatocellular injury than the commercially available suckling pig hepatocyte growth-promoting factor (PHGF). Conclusion: Danshensu has anti-TAA liver injury and stimulates the regeneration of hepatocytes after injury
