目的:观察基于“脑-肠轴”学说组方的清开通灌肠液,对内毒素性发热兔的退热效应及VIP、IL-2在发热中的变化。方法:日本大耳白家兔60只,随机分成4组:正常组、模型组、清开灵组和清开通组。模型组、清开灵组及清开通组,各组经耳缘静脉注射无热源生理盐水稀释的内毒素,建立兔内毒素性发热模型。同时清开灵组、清开通组分别予以清开灵、清开通灌肠液灌肠。注射内毒素2h后,清开灵组、清开通组再予灌肠1次。4h后,各组动物随机抽取10只,采集脑、肠及外周血标本,用放射免疫法检测脑、肠及血浆中VIP、IL-2含量。持续观察每组另外5只动物体温变化情况,每间隔10min记录1次体温,连续测温12h。结果:清开灵组和清开通组动物的体温与模型组相比都有明显降低(P<0.01),但清开通组降低比清开灵组更为显著(P<0.05)。脑肠及血浆中VIP和IL-2的含量都显著低于模型组(P<0.01),尤其清开通组较清开灵组降低幅度更大,二者有显著差异(P<0.05)。结论:“清开”与“通腑”合治,较单一清开法在退热及提高机体免疫方面效果更著,VIP和IL-2在发热中起正调节作用。
OBJECTIVE: To observe the antipyretic effect of Qingkai Tong enema and its effect on endotoxin-induced fever in rabbits and the change of VIP and IL-2 in fever based on the theory of “brain-gut axis”. Methods: 60 Japanese white rabbits were randomly divided into 4 groups: normal group, model group, Qingkailing group and Qingkailong group. Model group, Qingkailing group and Qingkailong group. The rabbits in each group were injected endotoxin without heat-normal saline into rabbits in each group to establish the endotoxin-induced fever model in rabbits. At the same time, Qingling Group, Qingkai Tong group were opened to clear Ling, clear open enema enema. After 2h injection of endotoxin, Qingkailing group, Qingkai Tong group again enema 1 times. After 4 hours, 10 animals were randomly selected from each group to collect brain, intestine and peripheral blood samples, and the levels of VIP and IL-2 in brain, intestine and plasma were detected by radioimmunoassay. Continue to observe the other five animals in each group changes in body temperature, temperature was recorded every 10min body temperature, continuous temperature 12h. Results: The body temperature of Qingkailing group and Qingkailong group were significantly lower than that of model group (P <0.01), but Qingkailong group decreased more significantly than Qingkailing group (P <0.05). The levels of VIP and IL-2 in brain, intestine and plasma were significantly lower than those in model group (P <0.01), especially in Qingkailong group than those in Qingkailing group. There was significant difference between the two groups (P <0.05). Conclusion: The combination of “Qingkai” and “Tongfu” can improve the immunity more than the single clearing method. VIP and IL-2 play a positive regulatory role in the fever.
