SD大鼠呼吸道染毒(1900±50)ppm氯乙烯2小时后,以肝脏灌流加酶消化方法获得肝实质性细胞和非实质性细胞,用淋巴细胞分离液分离外周血淋巴细胞,用单细胞微量凝胶电泳(MGE)技术测定上述细胞DNA损伤情况。结果发现,整体暴露氯乙烯可引起肝实质性和非实质性细胞的DNA损伤,但程度不同,且与体外试验结果相一致。氯乙烯还可引起外周血淋巴细胞DNA损伤。
Two hours after the exposure of SD rat to the airway (1900 ± 50) ppm of vinyl chloride, hepatic parenchymal cells and non-parenchymal cells were obtained by perfused liver and enzymatic digestion method. Peripheral blood lymphocytes were separated by lymphocyte separation liquid, Micro-gel electrophoresis (MGE) technique was used to determine DNA damage. The results showed that the overall exposure of vinyl chloride can cause DNA damage in liver parenchymal and non-parenchymal cells, but to a different degree, and consistent with in vitro test results. Vinyl chloride can also cause peripheral blood lymphocyte DNA damage.
