目的探讨1,2-二甲基-3-羟基-4-吡啶酮(DFP)对铝染毒雄性大鼠生殖损伤的保护作用。方法将35只健康清洁级Wistar雄性大鼠按体重随机分为5组,分别为阴性对照(生理盐水)组和铝单独染毒组以及铝+低、中、高剂量DFP组,每组7只,采用灌胃方式染毒,每天1次,每周染毒5 d,间隔2 d。前8周,铝染毒组给予三氯化铝195.9 mg/kg。后2周,铝单独染毒组给予生理盐水1 ml/d,铝+低、中、高剂量DFP组分别给予13.72、27.44和54.88 mg/kg的DFP溶液,每天1次。测定大鼠睾丸组织中乳酸脱氢酶(LDH)和β-葡糖苷酸酶(β-G)活力,分别统计外周血嗜多染红细胞微核细胞率和精子畸形率。结果与阴性对照组比较,铝单独染毒组大鼠睾丸组织中β-G活力显著降低(P<0.05);随着DFP给药剂量的升高,大鼠睾丸组织中β-G活力呈上升趋势;其中,铝+高剂量DFP组大鼠睾丸组织中β-G活力显著高于铝单独染毒组(P<0.05)。铝单独染毒组和铝+各剂量DFP组大鼠睾丸组织中LDH活力均显著低于阴性对照组(P<0.05);随着DFP给药剂量的升高,大鼠睾丸组织中LDH活力呈先下降后上升的趋势。随着DFP给药剂量的升高,外周血嗜多染红细胞微核率和精子畸形率均呈下降趋势,其中,铝+中、高剂量DFP组均显著低于铝单独染毒组(P<0.05)。结论 DFP能够恢复铝染毒导致的大鼠部分睾丸标志酶的活力,拮抗铝所致的遗传损伤,对生殖系统具有保护作用。
Objective To investigate the protective effects of 1,2-dimethyl-3-hydroxy-4-pyridone (DFP) on reproductive injury in aluminum-exposed male rats. Results Compared with the negative control group, the β-G activity in testis tissue of aluminum-exposed rats significantly decreased (P <0.05). With the increase of DFP dose, the β-G activity in testis tissue increased The β-G activity in testis of aluminum + high-dose DFP group was significantly higher than that of aluminum alone (P <0.05). The activity of LDH in testis of aluminum alone and aluminum + DFP group was significantly lower than that of negative control group (P <0.05). With the increase of DFP dose, LDH activity in testis tissue of rats After the first decline in the upward trend. The micronucleus rate and sperm abnormality rate of erythrocytes in peripheral blood decreased with the increase of DFP dose, and the levels of aluminum + medium and high dose DFP group were significantly lower than that of aluminum alone group (P < 0.05). Conclusion DFP can restore the vitality of some testis enzymes in rats induced by aluminum exposure and antagonize the genetic damage caused by aluminum and have a protective effect on the reproductive system.
