目的:观察不同浓度的甘草酸对内毒素引起的斑马鱼致命性感染的保护作用。方法:取发育正常的受精后4 d(4dpf)的斑马鱼幼鱼,经卵黄显微注射LPS(2 ng/条)建立内毒素感染模型。以不同浓度的甘草酸处理上述幼鱼,分别于处理后24h、48 h、72 h观察幼鱼的表型和死亡率的变化。结果:与模型组相比,24 h后不同浓度的甘草酸均能减轻斑马鱼的感染,降低死亡率(P<0.01);48 h后,模型组死亡率为100%,0.625 mg/ml甘草酸组死亡率为47.9%,1.25、2.5 mg/ml甘草酸组死亡率均低于23%;72 h后,0.625 mg/ml甘草酸组死亡率增加到59.37%,1.25 mg/ml甘草酸组死亡率为25.54%,2.5 mg/ml甘草酸组死亡率为19.61%。结论:甘草酸对内毒素引起的斑马鱼幼鱼致命性感染有明显的保护作用,且斑马鱼存活率与甘草酸的浓度正相关。
Objective: To observe the protective effect of different concentrations of glycyrrhizin on the deadly infection caused by endotoxin in zebrafish. Methods: The endotoxin infection model was established after 4 days (4dpf) normal fertilization of zebrafish embryos by LPS injection (2 ng / bar). The juveniles were treated with different concentrations of glycyrrhizic acid and the changes of phenotype and mortality of juvenile fish were observed at 24h, 48h and 72h after treatment. Results: Compared with the model group, glycyrrhizin at different concentrations could reduce the infection of zebrafish and reduce the mortality after 24 h (P <0.01). After 48 h, the mortality of the model group was 100% and 0.625 mg / The mortality rate of acid group was 47.9%, and the mortality rates of 1.25, 2.5 mg / ml glycyrrhizin group were all less than 23%. After 72 hours, the mortality of 0.625 mg / ml glycyrrhizin group increased to 59.37% and 1.25 mg / The mortality rate was 25.54%. The mortality rate of 2.5 mg / ml glycyrrhizin group was 19.61%. CONCLUSION: Glycyrrhizic acid has a significant protective effect on the lethal infection of endotoxin-induced zebrafish juveniles, and the survival rate of zebrafish is positively correlated with the concentration of glycyrrhizin.
