目的:探讨电刺激对脑梗死大鼠运动功能及骨形成蛋白2、6、7(BMP2,6,7)表达的影响。方法:健康雄性SD大鼠,制作大脑中动脉闭塞模型,将造模成功的大鼠随机分为对照组、电刺激组各48只,假手术组32只,正常组8只。对照组、电刺激组、于脑梗死后每天接受自然恢复、电刺激治疗6 h、12 h、24 h、3 d、7 d和21 d。并在上述时间点,各组取8只大鼠,进行走平衡木试验观察大鼠偏瘫肢体功能恢复情况,然后断头处死动物取脑,石蜡包埋切片行免疫组织化学染色,检测梗死灶周围皮质及正常大脑皮质BMP2,6,7的表达。结果:第7天、21天时,电刺激组肢体功能恢复较对照组明显(P<0.05)。正常组和假手术组BMP-2,6,7表达无统计学差异。对照组、电刺激组BMP-2表达均先下调,电刺激组大鼠BMP-2下调时间早于对照组,21 d恢复至正常水平,对照组、电刺激组BMP-6,7均上调,BMP-7于7 d灰度值上升,至21 d恢复至正常水平,BMP6则一直保持高水平,电刺激组BMP-6,7表达高于对照组。结论:电刺激组偏瘫肢体功能的恢复优于对照组,电刺激下调BMP-2表达,上调BMP-6,7表达。
Objective: To investigate the effects of electrical stimulation on motor function and the expression of bone morphogenetic protein 2,6,7 (BMP2,6,7) in rats with cerebral infarction. Methods: Male Sprague-Dawley rats were randomly divided into control group, 48 rats in each group, 32 sham operation group and 8 normal control group. The rats in the control group and the electrical stimulation group received natural recovery and electrical stimulation daily for 6 h, 12 h, 24 h, 3 d, 7 d and 21 d after cerebral infarction. At the above time point, 8 rats in each group were taken to take the balance wood test to observe the functional recovery of the hemiparesis limbs in rats. Then the animals were sacrificed and the brains were taken and the paraffin embedded sections were used for immunohistochemical staining to detect the cortex around the infarct And normal cerebral cortex BMP2,6,7 expression. Results: On the 7th day and 21th day, the functional recovery of the limb in the electrical stimulation group was more obvious than that in the control group (P <0.05). There was no significant difference in BMP-2,6,7 expression between normal group and sham operation group. The expression of BMP-2 in the control group and the electrical stimulation group were all down-regulated. The down-regulation time of BMP-2 in the electrical stimulation group was earlier than that in the control group, and returned to the normal level on the 21st day. The level of BMP-7 increased on the 7th day, returned to the normal level on the 21st day, BMP6 maintained a high level, and the expression of BMP-6,7 in the electro-stimulated group was higher than that of the control group. Conclusion: Electrical stimulation of hemiplegia limb function recovery is better than the control group, electrical stimulation down-regulate BMP-2 expression, up-regulate BMP-6,7 expression.
