目的研究瞬时受体电位香草醛亚家族1(TRPV1)在高脂处理肝细胞中的变化及其作用。方法将肝细胞(BRL-3A)分为对照组和250、500μmol·L~(-1)软脂酸钠组,加入辣椒素和辣椒卓平对肝细胞进行干预,通过Q-PCR法测量肝细胞中TRPV1的表达水平,并采用流式细胞仪检测肝细胞的凋亡水平。结果肝细胞的凋亡比例随软脂酸钠浓度的增加而增加,其浓度为500μmol·L~(-1)时,TRPV1的表达水平明显增加;辣椒素能够上调TRPV1的表达水平并减轻高脂所诱导的肝细胞凋亡;而辣椒卓平能够逆转辣椒素对肝细胞的保护作用。结论高脂能够激活肝细胞中的TRPV1通道,同时,上调TRPV1,减少高脂诱导的肝细胞凋亡。
Objective To study the changes of transient receptor potential vanilloid subfamily 1 (TRPV1) in hepatocytes treated with high fat diet and its effects. Methods Liver cells (BRL-3A) were divided into control group and 250, 500μmol·L -1 sodium palmitate group. Capsaicin and chilli peppers were added to intervene hepatocytes, and the liver was measured by Q-PCR The expression level of TRPV1 was detected by flow cytometry and the apoptosis of hepatocytes was detected by flow cytometry. Results The percentage of apoptotic hepatocytes increased with the increase of sodium palmitate concentration. The concentration of TRPV1 increased significantly at the concentration of 500 μmol·L -1. Capsaicin increased the expression of TRPV1 and decreased the expression of high lipid Induced hepatocellular apoptosis; and pepper Zhuo Ping can reverse the protective effect of capsaicin on liver cells. Conclusion High-fat can activate TRPV1 channel in hepatocytes, meanwhile, up-regulate TRPV1 and decrease hepatocyte apoptosis induced by high fat.
