根据GAMcDNA所编码的氨基酸序列合成了GAM分子羧基端23个氨基酸的多肽,并利用GMBS双功能试剂将人工合成多肽成功地与OVA进行了交联,免疫家兔制备了抗GAM合成多肽的抗体,并经亲和层析进行了纯化。对此抗体进行鉴定的结果表明,抗GAM合成多肽的抗体可与天然GAM分子发生特异性反应,并可用于免疫沉淀和Westernblot,为GAM分子功能的研究提供了有力的工具。
According to the amino acid sequence encoded by GAMcDNA, a polypeptide of 23 amino acids at the carboxyl terminus of GAM molecule was synthesized, and the synthetic peptide was successfully cross-linked with OVA using GMBS bifunctional reagent. Anti-GAM antibody was prepared by immunizing rabbits. And purified by affinity chromatography. The results showed that the anti-GAM antibody could react specifically with native GAM and could be used in immunoprecipitation and Western blotting, which provided a powerful tool for studying the function of GAM.
