目的探讨色素上皮衍生因子(PEDF)在早期宫颈鳞癌中的表达和临床意义。方法采用PV-9000免疫组织化学法对50例宫颈鳞癌(ICC)、18例宫颈上皮内瘤变(CIN)、35例正常宫颈组织(NCE)进行PEDF、CD34、Ki-67染色,采用半定量法计算PEDF的表达程度,并计算微血管密度(M VD)和癌细胞增殖指数(Ki-67标记)。结果 PEDF在NCE、CIN、ICC中阳性率分别为100.00%、77.78%和48.00%,差异有统计学意义(P<0.05)。PEDF在ICC中的低表达与盆腔淋巴结转移、脉管浸润有关(P<0.05),而与年龄、FIGO分期、肿瘤大小及分化程度无关(P>0.05)。PEDF在ICC中的表达与MVD(P<0.01,r=-0.642 4)以及Ki-67(P<0.01,r=-0.401 6)呈负相关。结论 PEDF表达下调可能在ICC血管增生、癌细胞增殖侵袭转移过程中起重要作用。
Objective To investigate the expression of pigment epithelium-derived factor (PEDF) in early cervical squamous cell carcinoma and its clinical significance. Methods Fifty cases of squamous cell carcinoma of the cervix (ICC), 18 cases of cervical intraepithelial neoplasia (CIN) and 35 cases of normal cervical tissue (NCE) were stained with PEDF, CD34 and Ki-67 by PV-9000 immunohistochemistry. Quantitative method was used to calculate the expression of PEDF, and to calculate the microvessel density (MVD) and the proliferation index of cancer cells (Ki-67 marker). Results The positive rates of PEDF in NCE, CIN and ICC were 100.00%, 77.78% and 48.00%, respectively, with significant difference (P <0.05). The low expression of PEDF in ICC was correlated with pelvic lymph node metastasis and vascular invasion (P <0.05), but not with age, FIGO stage, tumor size and differentiation (P> 0.05). PEDF expression in ICC was negatively correlated with MVD (P <0.01, r = -0.642 4) and Ki-67 (P <0.01, r = -0.401 6). Conclusion The down-regulation of PEDF expression may play an important role in the proliferation of ICC and the invasion and metastasis of cancer cells.
