目的 探讨Cyclin D1及MDM 2在胃粘膜癌变过程中的作用及其临床意义。方法 采用流式细胞术 (FCM)和免疫荧光技术对胃癌及癌前病变的细胞中Cyclin D1及MDM 2蛋白表达进行定量检测。结果 Cyclin D1、MDM 2蛋白表达出现在癌前病变 ,随病变加重 ,表达增强 ;在胃癌中显著高于癌前病变 (P <0 .0 1) ;且与胃癌的生物学行为有关。在胃粘膜不典型增生Ⅲ级中Cyclin D1的表达明显高于其它癌前病变(P <0 .0 5 )。结论 Cyclin D1、MDM 2基因在胃粘膜癌变过程中起作用 ,监测该基因的表达对胃癌的诊断、治疗及预后有临床意义。
Objective To investigate the role of Cyclin D1 and MDM 2 in the carcinogenesis of gastric mucosa and its clinical significance. Methods Flow cytometry (FCM) and immunofluorescence were used to detect the expression of Cyclin D1 and MDM 2 protein in gastric cancer and precancerous lesions. Results The expressions of Cyclin D1 and MDM 2 proteins in precancerous lesion were increased with the increase of lesion. The expression of Cyclin D1 and MDM 2 protein in gastric cancer was significantly higher than that in precancerous lesion (P <0.01). The expression of Cyclin D1 and MDM 2 was related to the biological behavior of gastric cancer. The expression of Cyclin D1 in grade Ⅲ gastric mucosa dysplasia was significantly higher than that in other precancerous lesions (P <0.05). Conclusion The expressions of Cyclin D1 and MDM 2 play an important role in the carcinogenesis of gastric mucosa. It is of clinical importance to monitor the expression of Cyclin D1 and MDM2 in the diagnosis, treatment and prognosis of gastric cancer.
