目的探讨回转器模拟失重对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)自噬的影响。方法体外培养的HUVEC,在回转器模拟失重条件下培养48 h,同时设l G静止培养为对照组。逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)和Western blot方法检测Beclin1 mRNA和蛋白的表达;透射电镜观察HUVEC超微结构。结果回转模拟失重后HUVEC Beclin1 mRNA及蛋白表达显著高于对照组;回转48 h后细胞呈现自噬性改变。结论回转模拟失重可以导致HUVEC自噬增加。
Objective To investigate the effect of gyrator simulated weightlessness on autophagy in human umbilical vein endothelial cells (HUVEC). Methods HUVECs cultured in vitro were cultured for 48 h under simulated weightlessness in a rotator, and l G stationary culture was set as the control group. The expression of Beclin1 mRNA and protein was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. The ultrastructure of HUVEC was observed by transmission electron microscope. Results The expression of Beclin1 mRNA and protein in HUVEC after swirling simulated weightlessness was significantly higher than that in control group. The autophagy changes were observed 48 h after swirling. Conclusion Rotational simulated weightlessness can lead to autophagy increase in HUVEC.
