目的在mRNA水平检测5株不同转移习性的人鼻咽癌细胞株中BRMS1基因表达的差别,为进一步证实BRMS1基因是否在人鼻咽癌细胞株中有抑制转移作用且抑制转移能力是否与该5种细胞株转移习性具有一致性,为课题组进行下一步组织学实验以及动物实验提供细胞理论基础。方法体外培养5株人鼻咽癌细胞株CNE-1、SUNE-1、SUNE-1-5-8F、CNE-2Z、SUNE-1-6-10B,分别提取该5株细胞株中mRNA,应用实时荧光定量PCR法检测该5株细胞株中BRMS1基因mRNA的表达水平是否存在差异。结果 BRMS1基因的mRNA在5株细胞株中均有表达,但表达水平存在差异。SUNE-1-6-10B表达量为标准值,SUNE-1-5-8F、SUNE-1、CNE-2Z、CNE-1中BRMS1 mRNA的相对表达量与标准值SUNE-1-6-10B表达量相比为标准值的(23±1)%、(49±2)%、(81±1)%、(86±2)%,较SUNE-1-6-10B细胞株中BRMS1 mRNA表达水平分别下降了77%、51%、19%、14%(P<0.05)。结论 BRMS1基因在5株不同转移习性人鼻咽癌细胞株中mRNA的表达水平具有差异,在高转移习性细胞株中表达量低,而在低转移习性细胞株中表达量高,且其mRNA表达量随着细胞株的转移能力的增加而下降,从以上结果说明在鼻咽癌转移过程中BRMS1基因可能起到抑制作用。
Objective To detect the difference of BRMS1 gene expression among 5 human nasopharyngeal carcinoma cell lines with different metastatic potential at the mRNA level. Real-time fluorescent quantitative PCR method was used to detect whether the expression level of BRMS1 mRNA in the five cell lines was different. Results The mRNA of BRMS1 gene was expressed in all five cell lines, but the expression level was different. The relative expression level of BRMS1 mRNA in SUNE-1-5-8F, SUNE-1, CNE-2Z, CNE-1 and SUNE-1-6-10B (23 ± 1)%, (49 ± 2)%, (81 ± 1)%, (86 ± 2)%, respectively, compared with the standard values of BRMS1 mRNA expression in SUNE-1-6-10B cells Decreased by 77%, 51%, 19% and 14% respectively (P <0.05).
