对取自湖南湘西自治州龙山县乌龙山3个不同洞穴的湘西盲高原鳅(Triplophysa xiangxiensis)群体82尾样本的线粒体细胞色素b(Cytb)基因序列和控制区(D-loop)序列进行了研究,分析了其遗传分化及遗传多样性。湘西盲高原鳅线粒体Cytb基因序列片段长1 140 bp,D-loop序列片段长934 bp,在D-loop中发现6个多态位点且均为单一变异位点,D-loop序列中检测到7个单倍体,其单倍型多样性指数(h)在0.083至0.282之间,其核苷酸多样性(pi)指数范围为0.000 09~0.000 32。3个采样群体的遗传多样性均较低。全部样本的Cytb序列均一致,不存在变异,4种碱基T、C、A、G含量分别是28.3%、28.6%、28.1%、15.0%,AT含量较高。D-loop的分子变异方差分析(AMOVA)结果显示湘西盲高原鳅遗传变异系数F ST=-0.008 9,总遗传变异中,各年度种群内变异为100.89%,年度种群间变异为-0.89%,变异主要来自年度种群内部。
The mitochondrial cytochrome b (Cytb) gene sequences and control region (D-loop) sequences from 82 populations of Triplophysa xiangxiensis population from three different caves in Wulongshan, Longshan County, Hunan Province were studied , Analyzed its genetic differentiation and genetic diversity.
