目的:研究骨髓基质细胞支持造血的分子基础。方法;应用免疫胶体金单标和双标记技术,在长期培养的骨髓基质细胞(HLTBMC)粘附层中,对各种造血调节有关的粘附蛋白进行了免疫电镜定位。结果:长期培养的骨髓基质细胞上均可显示各种粘附蛋白的表达,一些双标记证明各种粘附蛋白与某些细胞的特异性标记(vWF和Fb-sp)是共存的,表明在HLTBMC中的内皮细胞和成纤维细胞可合成和分泌各种粘附蛋白,这些粘附蛋白结合在基质细胞表面膜和细胞外基质上,结论:粘附蛋白与基质细胞分泌的各种生长因子一起,构成了一个支持造血的复杂网络,是基质细胞支持造血的分子基础。
Objective: To study the molecular basis of bone marrow stromal cells supporting hematopoiesis. Methods Immuno-microscopic localization of adherent proteins involved in various hematopoietic regulation in long-term adherence layers of bone marrow stromal cells (HLTBMCs) was performed using immunogold gold single and double labeling techniques. RESULTS: The expression of various adherent proteins could be shown on the long-term cultured bone marrow stromal cells. Some double-labeled coexistence of specific markers (vWF and Fb-sp) of some adherent proteins with some cells showed that at Endothelial cells and fibroblasts in HLTBMC synthesize and secrete various adhesion proteins that bind to the stromal cell surface membrane and the extracellular matrix, concluding that adhesion proteins bind with various growth factors secreted by stromal cells , Constitutes a complex network that supports hematopoiesis and is the molecular basis for stromal cells supporting hematopoiesis.
