目的 :从分子和蛋白水平揭示在急性肝功能衰竭 (AL F)时不同一氧化氮合酶 (NOS)诱导产生的一氧化氮 (NO)在肝、肺、肾脏和肠组织中表达的变化。方法 :雄性 Wistar大鼠 6 0只 ,随机分为 6组 :假手术 (SO)组、AL F 6 h组、AL F 12 h组、L - Arg组、L - NAME组、L - Arg+L - NAME组 ,每组 10只 ,L - Arg用量 30 0 m g/kg,L - NAME用量 30 mg/kg;用原位杂交和免疫组化方法检测肝、肺、肾组织 e NOS m RNA、i NOS m RNA表达和小肠、大肠 e NOS和 i NOS表达。结果 :AL F 6 h组肺 e NOSm RNA表达明显增加 ,肝、肺、肾 i NOS m RNA表达亦显著增加 ,而 AL F 12 h组则明显降低 (P<0 .0 5 )。应用 L - Arg在 AL F 6h组肝、肾 i NOS m RNA表达明显降低 ,而应用 L - NAME和 (或 )应用 L - Arg在 AL F 6 h组肺 e NOS m RNA及肝、肺、肾 i NOSm RNA表达均显著降低 (P<0 .0 5 ) ;AL F 6 h组小肠、大肠黏膜 i NOS表达显著增加 ,而 e NOS表达在 6 h、12 h组明显降低 ,尤其在 12 h组降低更明显 (P<0 .0 5 ) ;应用 L - Arg和 (或 )应用 L - NAME时 ,小肠、大肠黏膜 e NOS、i NOS表达均明显降低(P<0 .0 5 ) ;应用 L- NAME大肠黏膜 e NOS、i NOS表达均明显降低 (P<0 .0 5 )。结论 :AL F早期肺、肾组织 i NOS m RNA表达显著增加
OBJECTIVE: To reveal the changes in the expression of nitric oxide (NO) induced by different nitric oxide synthase (NOS) in liver, lung, kidney and intestine in acute liver failure (AL F) at molecular and protein levels. Methods: Sixty male Wistar rats were randomly divided into 6 groups: sham operation (SO) group, AL F 6 h group, AL F 12 h group, L - Arg group, L - NAME group, L - Arg + L - NAME group, 10 rats in each group, L - Arg dosage 30 mg / kg and L - NAME dosage 30 mg / kg. In situ hybridization and immunohistochemistry were used to detect eNOS mRNA in liver, lung and kidney NOS m RNA expression and eNOS and i NOS expression in small intestine, large intestine. Results: The expression of eNOSmRNA in ALF 6h group was significantly increased, and the expression of iNOS m RNA in liver, lung and kidney was also significantly increased in ALF 6h group compared with that in ALF 12h group (P <0.05). The expression of iNOS m RNA in liver and kidney of ALF 6h group was significantly lower than that of L - Arg group (P <0.05) (P <0.05). The expression of iNOS in small intestine and large intestine mucosa of ALF 6 h group was significantly increased, while the expression of e NOS in 6 h and 12 h group was significantly decreased, especially in 12 h group (P <0.05). When L - Arg and / or L - NAME was applied, the expression of eNOS and iNOS in small intestine and large intestine was significantly decreased (P <0.05) - NAME colorectal eNOS, i NOS expression were significantly lower (P <0. 05).
