目的:通过研究DNA结合分化抑制蛋白1(inhibitors of DNA binding-1,Id-1)和基质金属蛋白酶9(matrix metalloproteinase-9,MMP-9)在结直肠腺癌中,分析Id-1和MMP-9与结直肠腺癌进展的关系。方法 :采用免疫组织化学法、RT-PCR和Western blot检测Id-1和MMP-9在50例结直肠癌患者癌组织及50例正常组织中的表达情况,分析二者与各临床病理因素的相关性。结果:Id-1和MMP-9在结直肠癌组织中的阳性表达率分别为72.00%(36/50)和78.00%(39/50),而在正常结直肠组织中的阳性率为24.00%(12/50)和22.00%(11/50),差异均有统计学意义(P<0.01)。Id-1和MMP-9在癌组织中的表达呈正相关(r=0.422,P=0.002)。Id-1和MMP-9的表达与结直肠腺癌的浸润深度、TNM分期、肝转移、淋巴结转移和脉管浸润具有相关性(P均<0.05),而与患者的性别、年龄、肿瘤大小和分化程度无关(P均>0.05)。RT-PCR和Western blot检测结果与免疫组织化学检测结果基本一致。生存分析显示Id-1和MMP-9表达与结直肠癌患者预后关系密切,Id-1和MMP-9高表达提示预后不良。结论:Id-1和MMP-9在结直肠癌中高表达,其表达水平与结直肠腺癌的浸润、转移具有高度相关性,二者联合检测对判断结直肠癌的侵袭、转移及预后具有重要指导意义。
OBJECTIVE: To detect the expression of Id-1 and MMP in colorectal adenocarcinoma by studying the expression of inhibitors of DNA binding-1 (Id-1) and matrix metalloproteinase-9 -9 and the progression of colorectal adenocarcinoma. Methods: The expressions of Id-1 and MMP-9 in 50 cases of colorectal cancer and 50 cases of normal tissues were detected by immunohistochemistry, RT-PCR and Western blot. Results: The positive rates of Id-1 and MMP-9 in colorectal cancer tissues were 72.00% (36/50) and 78.00% (39/50) respectively, while those in normal colorectal tissues were 24.00% (12/50) and 22.00% (11/50), respectively, with significant difference (P <0.01). The expression of Id-1 and MMP-9 in cancer tissues was positively correlated (r = 0.422, P = 0.002). The expression of Id-1 and MMP-9 were correlated with the depth of invasion, TNM stage, hepatic metastasis, lymph node metastasis and vascular invasion in colorectal adenocarcinoma (all P <0.05), but not with the gender, age, tumor size And not related to the degree of differentiation (P> 0.05). The results of RT-PCR and Western blot were consistent with those of immunohistochemistry. Survival analysis showed that the expression of Id-1 and MMP-9 were closely related to the prognosis of patients with colorectal cancer. The high expression of Id-1 and MMP-9 suggested poor prognosis. Conclusions: Id-1 and MMP-9 are highly expressed in colorectal cancer. The expression of Id-1 and MMP-9 is highly correlated with infiltration and metastasis of colorectal adenocarcinoma. The combined detection of Id-1 and MMP-9 is of great importance in judging the invasion, metastasis and prognosis of colorectal cancer Guiding significance.
