目的:探讨新生鼠缺氧缺血性脑损伤中 c-fos 和 c-jun 表达的意义。方法:Wistar 大鼠18只分为两组,实验组12只,对照组6只,制备脑缺氧缺血动物模型,用 RT-PCR 和斑点杂交方法检测新生鼠缺氧缺血性脑损伤后 c-fos 和 c-jun 在神经细胞中的表达。结果:c-fos 和 c-jun的表达在不同时间段具有显著性差异(P<0.001),其中72小时组和96小时组的表达比24小时高,缺氧缺血后24~96小时,c-fos 和 c-jun 在躯体感觉皮质区,海马回 CA3区和基底节区持续表达,与对照组相比有统计学意义(P<0.01);c-fos 和 c-jun 的表达在不同脑区也具有显著性差异(P<0.01),海马区与基底节区大于皮质区。结论:c-fos 和 c-jun 的表达增加不仅是脑缺氧缺血的结果,而且可能参与了对神经细胞损害的过程。
Objective: To investigate the expression of c-fos and c-jun in neonatal rats with hypoxic-ischemic brain damage. Methods: Eighteen Wistar rats were divided into two groups: experimental group (n = 12) and control group (n = 6). The animal model of hypoxic-ischemic brain damage was established. The hypoxic-ischemic brain damage was detected by RT-PCR and dot blot hybridization Expression of c-fos and c-jun in neural cells. Results: The expressions of c-fos and c-jun were significantly different at different time points (P <0.001). The expressions of c-fos and c-jun were higher than those of 24 hours and 72 hours and 96 hours, The expressions of c-fos and c-jun in the somatosensory cortex, hippocampus CA3 and basal ganglia were significantly higher than those in the control group (P <0.01) The brain area also had significant difference (P <0.01), hippocampus and basal ganglia were larger than the cortex. Conclusion: The increased expression of c-fos and c-jun is not only the result of hypoxic-ischemic brain damage, but also may be involved in the process of nerve cell damage.
