研究了2-溴乙酰芳胺对邻氨基苯甲醇的烷基化反应,反应主要发生在N原子上,并由此合成了一系列新型芳胺基取代乙酰芳胺类化合物(4a~4e)。对反应溶剂、碱的种类及用量、反应温度、时间、反应物的量之比等条件对反应的影响进行了研究,获得优化反应条件。在优化条件下化合物4a~4e的收率为68.9%~72.8%。对反应影响比较大的因素主要是溶剂,采用v(N,N-二甲基甲酰胺)∶v(四氢呋喃)=1∶2的混合溶剂可使反应收率从36.1%(THF中)提高到70.3%。用IR、1H NMR和13C NMR对化合物4a~4e的结构进行了分析与表征。
The alkylation reaction of 2-bromoacetyl aromatic amine with o-aminobenzyl alcohol was studied. The reaction occurred mainly on the N atom, and a series of novel arylamino-substituted acetylaryl amines (4a ~ 4e) were synthesized. The influence of the reaction solvent, the type and amount of alkali, the reaction temperature, the time and the ratio of reactants on the reaction was studied, and the optimal reaction conditions were obtained. Under optimized conditions, the yields of compounds 4a ~ 4e ranged from 68.9% to 72.8%. The main influential factors on the reaction were solvent, and the reaction yield increased from 36.1% (THF) to (1: 2) v (N, N-dimethylformamide) 70.3%. The structures of compounds 4a ~ 4e were analyzed and characterized by IR, 1H NMR and 13C NMR.
