以美国红枫腋芽茎段为外植体,采用组织培养方法,研究了不同激素配比对美国红枫无菌体系的建立、启动、增殖、壮苗培养、试管苗生根移栽的影响。结果表明:带腋芽茎段最佳消毒方式为以70%酒精浸泡30s,1%升汞消毒6min,成活率和抽芽率最高,分别达到75.22%和95.13%;NAA对启动培养的影响大于6-BA,接种于培养基MS+NAA 0.05mg·L~(-1)+6-BA0.5mg·L~(-1)中的外植体诱导率最高,为62.80%,培养基及激素的最佳配比为NN69+2,4-D0.1mg·L~(-1)+TDZ 0.3mg·L~(-1),诱导率达到80.21%;增殖培养阶段的最佳培养基为NN69+NAA 0.1mg·L~(-1)+TDZ 0.3mg·L~(-1),增殖系数为4.2;MS+2,4-D 0.1mg·L~(-1)+6-BA0.5mg·L~(-1)能有效促进小苗的生长;生根培养最适培养基MS+NAA 0.1mg·L~(-1)+2,4-D0.5mg·L~(-1),生根率达99.8%,生根条数达7.9,根长达6.5cm;练苗后移栽到蛭石+珍珠岩(1∶1)基质中,移栽成活率为75.2%。
In this study, the tissue culture method was used to study the effects of different hormone combinations on establishment, initiation, proliferation, seedling culture and rooting and transplanting of Rhodiola salsa. The results showed that the best way of disinfecting stem with axillary buds was 75s2% and 95.13%, respectively, with 70% alcohol soaked for 30s and 1% mercuric chloride for 6min. The optimal culture medium for rooting was MS + NAA 0.1 mg · L -1 and 2,4-D 0.5 mg · L -1, the rooting rate was 99.8 %, The rooting number of 7.9, the root length of 6.5cm; After transplanting to vermiculite / perlite (1: 1) substrate after seedling cultivation, the survival rate of transplanting was 75.2%.
