目的:观察黄芪多糖(astragalus polysaccharide,APS)对高血压病血瘀证患者血清损伤的人脐静脉内皮细胞(HUVEC-C)形态学、活性的影响。方法:将培养的高血压病血瘀证患者血清损伤的HUVEC-C分别用不同浓度的APS含药血清干预后,倒置显微镜下观察其形态学变化,MTT法检测其活性。结果:APS呈剂量依赖性升高损伤的细胞OD值,且随着药物组浓度的增加,细胞的形态趋向正常细胞。结论:APS能够抵抗高血压病血瘀证患者血清中有害因子对HUVEC-C的损伤作用,能增强细胞的活性和维持细胞的形态结构,并呈剂量依赖性。
OBJECTIVE: To observe the effect of astragalus polysaccharide (APS) on the morphology and activity of human umbilical vein endothelial cells (HUVEC-C) in patients with hypertension and blood stasis syndrome. Methods: HUVEC-C with serum-injured HUVEC-C in patients with blood-stasis syndrome was cultured in different concentrations of APS-containing serum. The morphological changes were observed under an inverted microscope. The activity of HUVEC-C was detected by MTT assay. Results: APS showed a dose-dependent increase in the OD value of injured cells, and with the increase of drug concentration, the cell morphology tended to normal cells. Conclusion: APS can resist the damage of HUVEC-C by the serum harmful factors in patients with blood-stasis syndrome, enhance the activity of cells and maintain the morphological structure of cells in a dose-dependent manner.
