目的研究家蝇幼虫血淋巴纯化蛋白(anticancer protein from Musca domestica larva hemolymph,MAC-1)对体外培养的人宫颈癌Hela细胞抑制作用引起的形态学改变。方法应用四甲基偶氮噻唑蓝(MTT)法、光学显微镜、HE和吖啶橙(AO)染色、透射电镜(SEM)的方法检测MAC-1对体外培养的人宫颈癌Hela细胞的抑制作用及形态学改变。结果 MAC-1对人宫颈癌Hela细胞株的增殖抑制作用呈剂量依赖性,同时呈时间依赖性。MAC-112.5μg/ml作用12 h后在倒置显微镜、HE及AO染色和透射电镜均观察到典型细胞凋亡形态变化。结论 MAC-1对人宫颈癌Hela细胞株具有抑制增殖及诱导凋亡作用。
Objective To study the morphological changes induced by the inhibitory effect of Musca domestica larva hemolymph (MAC-1) on human cervical cancer Hela cells cultured in vitro. Methods The inhibitory effects of MAC-1 on human cervical carcinoma Hela cells were detected by MTT method, light microscope, HE and acridine orange (AO) staining and transmission electron microscopy (SEM) And morphological changes. Results The inhibitory effect of MAC-1 on human cervical cancer Hela cell line was dose-dependent and time-dependent. MAC-112.5μg / ml for 12 h after the inverted microscope, HE and AO staining and transmission electron microscopy were observed typical apoptotic morphological changes. Conclusion MAC-1 can inhibit proliferation and induce apoptosis of human cervical cancer Hela cell line.
