目的分析妊娠期高血压疾病孕妇与正常孕妇血浆中microRNA-19a(miR-19a)的差异性表达特征,并探讨其临床应用。方法选择2013年1月至2015年6月泸州医学院附属医院收治的妊娠期高血压疾病孕妇90例作为妊娠期高血压疾病组,按孕妇病情再分为妊娠期高血压组(30例)、轻度子痫前期组(30例)、重度子痫前期组(30例)。另选择同期在本院行孕检的30例健康孕妇作为正常组,应用实时荧光定量聚合酶链式反应测定不同组孕妇血浆中miR-19a的相对表达量,比较妊娠期高血压疾病组与正常组孕妇miR-19a的表达差异,并分类统计比较妊娠期高血压疾病各亚组miR-19a表达量,评估miR-19a的临床诊断应用价值。结果与正常组比较,妊娠期高血压疾病组miR-19a表达量显著升高(P<0.05);不同妊娠期高血压疾病亚组miR-19a表达量比较,重度子痫前期组miR-19a表达量最高,其后依次为轻度子痫前期组、妊娠期高血压组、正常组(P<0.05)。结论妊娠期高血压疾病孕妇与正常孕妇血浆中miR-19a表达量存在差异性,妊娠期高血压疾病孕妇血浆中存在升高趋势,对于妊娠期高血压疾病的临床诊断具有一定指导意义。
Objective To analyze the differential expression of microRNA-19a (miR-19a) in plasma of pregnant women with gestational hypertension and normal pregnant women and to explore its clinical application. Methods Ninety pregnant women with gestational hypertension admitted to the Affiliated Hospital of Luzhou Medical College from January 2013 to June 2015 were enrolled as hypertensive disorder complicating pregnancy. The pregnant women were further divided into three groups according to the pregnant women ’s condition: gestational hypertension group (n = 30) Mild preeclampsia group (30 cases) and severe preeclampsia group (30 cases). Another 30 healthy pregnant women undergoing pregnancy test at the same period in our hospital were selected as the normal group. The relative expression of miR-19a in different groups of pregnant women was measured by real-time fluorescence quantitative polymerase chain reaction. Compared with the normal MiR-19a expression in pregnant women group, and statistical comparison of subgroup miR-19a expression level in hypertensive disorders of pregnancy to evaluate the value of miR-19a clinical diagnosis. Results Compared with the normal control group, the expression of miR-19a in hypertensive disorder complicating pregnancy was significantly increased (P <0.05). The expression of miR-19a in subgroup of hypertensive disorder complicating pregnancy was significantly higher than that in normal group The highest level was followed by mild preeclampsia group, gestational hypertension group and normal group (P <0.05). Conclusions The expression of miR-19a in pregnant women with gestational hypertension is different from that in normal pregnant women. There is an increasing tendency in the plasma of pregnant women with hypertensive disorder complicating pregnancy, which is of guiding significance in the clinical diagnosis of hypertensive disorder complicating pregnancy.
