铅对仔鼠脑Brn-3a蛋白及神经丝蛋白表达影响

目的探讨铅对中枢神经系统神经细胞内转录因子Brn-3a及神经丝蛋白(NF)表达的影响。方法雌性大鼠在围产期经饮水染醋酸铅(对照组蒸馏水、低0.5,中1.0,高2.0g/L染毒后,取其21d龄仔鼠脑组织,采用免疫组织化学方法对Brn-3a蛋白及神经丝蛋白进行检测,以了解其表达水平。结果免疫组织化学图像分析表明,各染铅组脑组织皮层、海马及小脑的Brn-3a蛋白表达的阳性面积比[Aa(%)]、平均灰度与对照组比较,差异有统计学意义(P<0.05或P<0.01),并呈剂量依赖关系。染铅组Brn-3a蛋白表达低于对照组。与对照组比较,低铅组、中铅组NF蛋白阳性细胞无明显改变,而高铅组大脑皮层、海马和小脑NF蛋白阳性细胞普遍数量减少,着色变浅,神经丝蛋白表达减少。结论在染铅后,大鼠脑组织Brn-3a和NF蛋白表达水平有所下降,提示铅干扰了Brn-3a蛋白的正常表达模式,Brn-3a作为转录调节因子进而影响下游NF蛋白的正常表达。 Objective To investigate the effects of lead on the expression of Brn-3a and NF in neural cells of the central nervous system. Methods Female rats were exposed to lead acetate for drinking during the perinatal period (control group distilled water, low 0.5, medium 1.0, high 2.0g / L after taking the brain tissue of 21 d old rats, immunohistochemistry Brn-3a Protein and neurofilament were detected in order to understand its expression level.Results Immunohistochemical image analysis showed that the positive area ratio [Aa (%)] of Brn-3a protein expression in brain tissue cortex, hippocampus and cerebellum of each lead group, Compared with the control group, the average gray level was significantly different (P <0.05 or P <0.01), and the expression of Brn-3a protein in the lead-exposed group was lower than that of the control group , While there was no significant change in NF-positive cells in the lead group, while the prevalence of NF-positive cells in the hippocampus and cerebellum of the lead group was decreased, the staining was lighter and the expression of neurofilament protein was decreased.Conclusion After lead exposure, Brn-3a and NF protein expression decreased, suggesting that lead interfere with the normal expression pattern of Brn-3a protein, Brn-3a as a transcriptional regulator and thus affect the downstream normal NF protein expression.
