目的:了解新维甲类化合物联合γ干扰素(IFN-γ)对U937细胞的增殖分化作用及其机制。方法:检测U937细胞NBT阳性率、细胞表面CD14和CD68表达、c-myc和细胞周期变化,研究新维甲类化合物R9158和YS904012分别联合IFN-γ对U937细胞增殖分化的作用。结果:先用维甲类化合物作用24小时后换用IFN-γ诱导U937细胞分化,有明显的协同作用;若用相反顺序处理,它们的协同作用明显降低。新维甲类化合物24小时内能使细胞积聚于G0/G1期,下调c-myc基因。YS904012活性稍强。结论:新维甲类联合IFN-γ有协同诱导U937细胞分化的作用。维甲类化合物主要作用于细胞的早期阶段;IFN-γ主要在后期起作用。早期应用维甲类化合物在促进细胞分化中具有重要的作用。
OBJECTIVE: To investigate the proliferation and differentiation of U937 cells induced by neo-viridae combined with interferon-γ (IFN-γ) and its mechanism. Methods: The positive rate of NBT, the expression of CD14 and CD68 on the surface of U937 cells, c-myc and cell cycle were detected to study the effect of the new retinoid R9158 and YS904012 combined with IFN-γ on the proliferation and differentiation of U937 cells. Results: Synergistic effect was observed when IFN-γ was used to induce differentiation of U937 cells after treated with retinoids for 24 hours. Synergistic effect was observed when treated with the reverse order. The new retinoids can make cells accumulate in G0 / G1 phase and down-regulate c-myc gene within 24 hours. YS904012 activity slightly stronger. Conclusion: The combination of new dimension A and IFN-γ can synergistically induce the differentiation of U937 cells. The retinoid mainly acts on the early stages of the cell; IFN-γ mainly plays a role in the later stage. Early application of retinoids in promoting cell differentiation has an important role.
