目的:观测兔肾脏缺血-再灌注损伤(IRI)后血清白细胞介素-6(IL-6)动态变化及病理改变,探讨其潜在临床意义。方法成年新西兰大白兔10只用无创性动脉夹夹闭左肾肾蒂60 min制作肾IRI损伤模型,分别于IRI前、IRI后12 h、24 h、48 h抽取耳缘静脉血进行血清IL-6生化定量分析,并观察IRI肾形态学及组织病理学变化。结果(1)建模前、IRI-12h、IRI-24h及IRI-48h的血清IL-6均值分别为2.52±0.40μg/L、3.29±0.50μg/L、3.80±0.62μg/L和2.12±0.24μg/L,4组数据之间差异有统计学意义(F=33.56,P<0.01);(2)IRI-12h见近曲小管上皮细胞肿胀,空泡变性;IRI-24h细胞变性加重,间质有炎性细胞浸润,管周血管明显扩张淤血。IRI-48h病变程度均较IRI-24h减轻。结论兔肾IRI后血清IL-6浓度可间接评估IRI的严重程度。“,”Objective To observe the dynamic changes of serum IL-6 and pathological changes in rabbit renal ischemia reperfusion injury (IRI), and to explore its potential clinical significance. Methods 10 adult New Zealand white rabbits were used for renal IRI injury model by blocked the left renal pedicle using noninvasive arterial clip. The serum IL-6 were biochemical analyzed before IRI and 12, 24 and 48 hours after IRI with ear vein blood serum, respectively. The morphological and pathological changes of IRI kidney were observed. Results (1)The mean value of IL-6 was 2.52±0.40μg/L before IRI, 3.29±0.50μg/L in group IRI-12h, 3.80±0.62μg/L in group IRI-24h and 2.12±0.24μg/L in group IRI-48h, respectively. There were significant differences between four groups (F=33.56, P<0.01). (2)Group IRI-12h showed proximal tubular epithelial cell swelling with vacuolar degeneration. Group IRI-24h displayed the cell degeneration aggravated, interstitial infiltration of inflammatory cells and peritubular blood vessels dilated. The severity of group IRI-48h was alleviated than group IRI-24h. Conclusion The concentration of serum IL-6 may be used to evaluate the severity of IRI in rabbit renal IRI indirectly.
