目的建立毛连菜总酚的含量测定方法,并优选其大孔吸附树脂纯化工艺。方法采用紫外-可见分光光度法测定毛连菜总酚的含量,以吸附量、解吸率、解吸量等指标优选毛连菜总酚的大孔吸附树脂富集纯化工艺。结果所建立的质量控制方法的精密度、重复性和稳定性良好,其RSD均<3.0%。优选出了HP-20树脂对毛连菜总酚有较好的吸附、解吸性能,最佳工艺:上样的毛连菜水溶液总酚浓度为8.54 mg·m L?1,上样体积为1 BV,控制流速为2 BV·h-1。先用水和10%乙醇洗去杂质,用量各4 BV,收集4 BV的30%乙醇和3 BV的50%乙醇洗脱液,浓缩,即得。结论本实验建立的质量控制方法简单、准确,经HP-20树脂分离纯化,毛连菜总酚的纯度可达87%以上,且工艺稳定。
OBJECTIVE To establish a method for the determination of total phenols in Rumex gmelini and optimize the purification process of macroporous resin. Methods The content of total phenols was determined by UV - Vis spectrophotometry. The macroporous resin was used to enrich and purify total phenols with the indexes of adsorption, desorption and desorption. Results The quality control method established by the method had good precision, repeatability and stability with RSD <3.0%. The best adsorption and desorption properties of HP-20 resin on total phenolics of Broccoli were optimized. The optimum conditions were as follows: the total phenolic concentration of the sample was 8.54 mg · m L · 1 and the sample volume was 1 BV, the control flow rate of 2 BV · h-1. First with water and 10% ethanol wash impurities, the amount of 4 BV, 4 BV of 30% ethanol and 3 BV of 50% ethanol eluent, concentrated, that is, too. Conclusion The method of quality control established in this experiment is simple and accurate. The purity of total phenols in the hairy broccoli can be up to 87% with the separation and purification by HP-20 resin, and the process is stable.
