目的:建立用酶联免疫(ELISA)法测定去氨加压素血药浓度的快速、简便方法,并研究其药动学。方法:用人去氨加压素(dDAVP)ELISA试剂盒对8例受试者给药后各时间段的血样进行血管加压素含量测定,用SPSS 18.0对检测平均值作药时曲线图并计算药动学相关参数。结果:ELISA法测定去氨加压素标准曲线回归方程为:Y=92.364 X+9.655 5,R2=0.998 1,平均回收率为100.96%;t1/2=(81.1±10.9)min;Cmax=(175.1±8.8)pg·mL-1;AUC0→200=(18 415±964)pg·h·mL-1;ClT=(52.8±2.6)mL·min-1。结论:ELISA法用于测定去氨加压素血药浓度及药动学研究的方法快速方便,结果稳定可靠。
Objective: To establish a rapid and simple method for the determination of plasma concentration of desmopressin by enzyme-linked immunosorbent assay (ELISA) and to study its pharmacokinetics. Methods: The vasopressin contents of blood samples of 8 subjects after dosing were determined by dDAVP ELISA kit. The pharmacokinetic parameters were calculated by SPSS 18.0 Pharmacokinetic parameters. Results: The regression equation of standard curve of desmopressin by ELISA was Y = 92.364 X + 9.655 5, R2 = 0.998 1, the average recovery was 100.96%; t1 / 2 = (81.1 ± 10.9) min; Cmax = 175.1 ± 8.8) pg · mL -1; AUC0 → 200 = (18 415 ± 964) pg · h · mL -1; ClT = (52.8 ± 2.6) mL · min -1. Conclusion: ELISA method for the determination of desmopressin plasma concentration and pharmacokinetic methods quickly and easily, the results are stable and reliable.
