目的:观察成纤维细胞生长因子5 (FGF5)对人胰腺癌细胞生长和肝转移能力的影响。方法:利用高表达FGF5的人胰腺癌PANC-1细胞系(北京大学第一医院外科大肠癌实验室提供),通过短发卡RNA(shRNA)稳定转染技术,构建FGF5低表达细胞系PANC-1-FGF5n -,并以未转染质粒和转染对照粒的PANC-1细胞作为对照。通过皮下注射各组细胞建立裸鼠皮下种植瘤模型;通过脾脏内注射各组细胞建立肝转移模型,6周后切取皮下种植瘤体并称重,切取肝转移标本,计数肝转移灶数目。单因素方差分析比较各组裸鼠皮下种植瘤质量和肝转移灶个数的差异,Spearman等级相关性分析各组裸鼠皮下种植瘤质量和肝转移灶个数与各组细胞FGF5表达水平之间的相关性。n 结果:蛋白质印迹法(Western blot)法证实FGF5低表达及对照质粒细胞构建成功;PANC-1-FGF5n -组皮下种植瘤质量[(0.42±0.19) g]明显低于Pan-1组[(1.35±0.45) g]和PANC-1-Vector组[(1.32±0.35) g],差异有统计学意义(n F=18.506,n P<0.01);PANC-1-FGF5n -组肝转移灶数目[(7.50±4.11)个]明显少于PANC-1组[(26.25±9.78)个]和PANC-1-Vector组[(26.25±9.78)个],差异有统计学意义(n F=12.971,n P<0.01);种植瘤质量和肝转移灶数与FGF5的表达量成正相关(n r=0.818、0.806,n P<0.01)。n 结论:FGF5在裸鼠体内能够促进胰腺癌细胞的生长和肝转移能力。“,”Objective:To explore the effects of the fibroblast growth factor 5 (FGF5) on the growth and the hematogenous metastasis of the human pancreatic cancer cells (PANC-1 cells).Methods:PANC-1 cells were stably transfected with FGF5 short hairpin RNA (shRNA) and contrast vector, respectively. Transfected cells were selected with G418. Cells from each group (PANC-1, PANC-1-Vector and PANC-1-FGF5n - cells) were inoculated subcutaneously into 8 nude mice (bagg′s albino, BALB/c nu/nu) respectively. The subcutaneous tumors were harvested and weighed. Cells from each group were injected into spleens of 8 nude mice by operations. Livers were harvested 6 weeks after nude mice liver metastasis model was constructed. Then the liver metastatic amounts were countered macroscopically. The quality of subcutaneous tumor and the number of liver metastases in each group were compared by one way analysis of variance (ANOVA), and the correlation between the quality of subcutaneous tumor and the number of liver metastases and the expression level of FGF5 was analyzed by Spearman correlation analysis.n Results:Western blotting showed that FGF5 shRNA and contrast vector cell lines were constructed successfully. The quality of the subcutaneous tumors of PANC-1-FGF5-cells [(0.42±0.19) g]was less than PANC-1 cells [(1.35±0.45) g] and PANC-1-Vector cells [(1.32±0.35) g], and the difference was significant (n F=18.506, n P<0.01). The liver metastatic amounts of PANC-1-FGF5n - cells (7.50±4.11) were less than PANC-1 cells (26.25±9.78) and PANC-1-Vector cells (26.25±9.78), and the difference was significant (n F=12.971, n P<0.01). The expression of FGF5 was highly positively correlated with the quality of the tumors and liver metastatic amounts (n r=0.818, 0.806, n P<0.01).n Conclusion:FGF5 could promote the growth and the liver metastasis of PANC-1 cells n in vivo in nude mice.n
