目的 建立对患者体内病毒水平进行客观定量检测的方法。方法 采用差示逆转录 -聚合酶链反应 ,共同扩增柯萨奇B组病毒 (CVB)目的基因和参照模板Mmyogene ,建立一种检测CVB复制水平的定量方法 ,并对苦瓜素体外抗CVB3感染疗效进行初步评价。结果 该方法成功检测到CVB3RNA水平 ,且苦瓜素处理组CVB3RNA水平明显低于对照组。结论 该方法具有简便、快速、可靠、重复性好等优点 ,同时检测多份样本 ,非常适合于大样本药物筛选 ,可为抗CVB病毒感染疗效的评价提供一个有效指标
Objective To establish a method for objective and quantitative detection of virus in patients. Methods Differential reverse transcriptase-polymerase chain reaction (PCR) was used to amplify the target gene of Coxsackie B virus (CVB) and the reference template Mmyogene to establish a quantitative method for detecting the replication level of CVB. The in vitro anti-CVB3 infection The efficacy of a preliminary evaluation. Results The method successfully detected CVB3RNA levels, and the levels of CVB3RNA in the control group were significantly lower than those in the control group. Conclusion The method is simple, rapid, reliable and reproducible. It can detect multiple samples simultaneously and is very suitable for drug screening of large samples. It can provide an effective index for evaluating the curative effect of CVB virus infection
