准噶尔乌头中一株苹果斑点落叶病菌拮抗菌的分离及鉴定

从准噶尔乌头(Aconitum soongaricum Stapf)的根茎中分离出1株对苹果斑点落叶病菌(Alternaria mali)有拮抗作用的内生菌XJAS-ZB-14,其主要生理生化指标与短小芽孢杆菌相符。利用16SrDNA通用引物对其基因组DNA进行扩增,并测序得到XJAS-ZB-14的部分16SrDNA序列,GenBank注册号为FJ237280。经Blastn比对,用软件MEGA4.0按照Neighbor-Joining方法构建16SrDNA系统发育树。与菌株XJAS-ZB-14同源性最高的菌株为Bacillus pumilus DSM27T(AY456263),相似性为100%,(G+C)mol%为53.41%,最终鉴定为短小芽孢杆菌Bacilluspumilus,并对其生长条件作了研究。 A strain of endophyte XJAS-ZB-14 with antagonistic activity against Alternaria mali was isolated from the rhizomes of Aconitum soongaricum Stapf. Its main physiological and biochemical indexes were consistent with those of B. pumilus. Genomic DNA was amplified by 16S rDNA universal primers and sequenced to obtain a partial 16S rDNA sequence of XJAS-ZB-14. The GenBank accession number was FJ237280. By Blastn alignment, the 16S rDNA phylogenetic tree was constructed using the software MEGA4.0 according to the Neighbor-Joining method. The strain with the highest homology with strain XJAS-ZB-14 was Bacillus pumilus DSM27T (AY456263) with a similarity of 100% and a (G + C) mol% of 53.41%, which was finally identified as Bacillus pumilus and its growth Conditions were studied.
