目的研究复方通痹胶囊的镇痛和抗类风湿关节炎活性,并对其作用机制进行初步研究。方法采用小鼠醋酸和热板致痛模型,研究复方通痹胶囊的镇痛活性;采用二甲苯耳肿、大鼠角叉菜胶足肿、棉球肉芽肿和佐剂性关节炎模型综合评价复方通痹胶囊对类风湿关节炎的影响;ELISA法检测佐剂性关节炎大鼠血清PGE_2水平。结果与溶媒对照组比较,复方通痹胶囊可明显减少醋酸所致小鼠扭体反应(P<0.01),明显延长热板所致小鼠痛阈值(P<0.05);与溶媒对照组比较,复方通痹胶囊可明显抑制角叉菜胶所致大鼠足肿胀(P<0.05),明显抑制大鼠棉球肉芽组织增生(P<0.01),但其对二甲苯所致小鼠耳肿无明显抑制作用(P>0.05);与溶媒对照组比较,复方通痹胶囊可明显减轻佐剂关节炎大鼠继发性足肿(P<0.05),降低大鼠关节炎指数(P<0.05),并可明显降低关节炎大鼠血清PGE_2水平(P<0.01)。结论复方通痹胶囊具有明显的镇痛和抗类风湿关节炎活性,其活性可能与抑制炎症大鼠PGE_2的生成有关。
Objective To study the analgesic and anti-rheumatoid arthritis activities of Fufang Tongbi Capsule and its mechanism of action. Methods The analgesic activity of Tongfang Tongbi Capsule was induced by acetic acid and hot plate in mice. The effects of compound Tongfang capsule on the analgesic activity of compound Fangbi Capsule were evaluated using xylene ear, rat carrageenan, granuloma and adjuvant arthritis Tongbi capsule on rheumatoid arthritis; ELISA assay of adjuvant arthritis in rats serum PGE_2 levels. Results Compared with the vehicle control group, Tongfang Tongbi Capsule could significantly reduce the writhing response induced by acetic acid (P <0.01) and prolong the pain threshold of mice induced by hot plate significantly (P <0.05). Compared with the vehicle control group, Tongfang capsule can significantly inhibit carrageenan-induced foot swelling in rats (P <0.05), significantly inhibited granulation tissue granulation tissue hyperplasia (P <0.01), but its xylene-induced mice ear swelling (P0.05) .Compared with the vehicle control group, Tongfang Tongbi Capsule could significantly reduce the secondary tumors of the adjuvant arthritis rats (P <0.05) and reduce the arthritis index (P0.05) , And can significantly reduce the level of serum PGE_2 in arthritic rats (P <0.01). Conclusion Fufang Tongbi Capsule has obvious analgesic and anti-rheumatoid arthritis activity, and its activity may be related to inhibiting the formation of PGE_2 in inflammatory rats.
