目的探讨白血病干细胞(leukemia stem cell,LSC)相关抗原在不同亚型急性白血病细胞中的表达规律。方法采用流式细胞术检测LSC相关抗原CD96,CD90,CD123,CD71等在50例不同亚型急性白血病细胞中的表达,包括急性粒细胞白血病未分化型(M1)、急性粒细胞白血病部分分化型(M2)、急性早幼粒细胞白血病(M3)、急性粒-单核细胞白血病(M4)和急性B淋巴细胞白血病。结果 CD96在M3的表达率(90.00%)明显高于M2(18.18%)和急性B淋巴细胞白血病(20.00%)(P<0.05);各亚型急性白血病均表达CD123,但差异无统计学意义(P>0.05);CD71在急性髓细胞白血病各亚型中(M1、M2、M3和M4)阳性表达率分别为80.00%、72.73%、90.00%和100.00%,明显高于急性B淋巴细胞白血病(P<0.05);CD90在急性B淋巴细胞白血病中阳性表达率为13.33%,高于急性髓细胞白血病(P>0.05)。结论 CD71与CD96的表达有亚型特异性,CD96可能具有指示系列分化和细胞分化程度的作用,CD71可用于区分急性淋巴细胞白血病和急性髓细胞白血病。
Objective To investigate the expression of leukemia stem cell (LSC) related antigen in different subtypes of acute leukemia cells. Methods Flow cytometry was used to detect the expression of CD96, CD90, CD123 and CD71 in 50 different subtypes of acute leukemia cells including acute myeloid leukemia undifferentiated (M1), partially differentiated acute myeloid leukemia (M2), acute promyelocytic leukemia (M3), acute myelomonocytic leukemia (M4) and acute B-lymphoblastic leukemia. Results The expression of CD96 in M3 was significantly higher than that in M2 (18.18%) and acute lymphoblastic leukemia (20.00%) (P <0.05). CD123 was expressed in all subtypes of acute leukemia, but the difference was not statistically significant (P> 0.05). The positive expression rates of CD71 in all subtypes of acute myeloid leukemia (M1, M2, M3 and M4) were 80.00%, 72.73%, 90.00% and 100.00%, respectively, which were significantly higher than those in acute B lymphoblastic leukemia (P <0.05). The positive rate of CD90 in acute B lymphoblastic leukemia was 13.33%, higher than that in acute myeloid leukemia (P> 0.05). Conclusion The expression of CD71 and CD96 is subtype-specific. CD96 may play a role in the differentiation and differentiation of cells. CD71 can be used to distinguish acute lymphoblastic leukemia from acute myeloid leukemia.
