以黑果花楸组培苗为试材,采用石蜡切片和叶表皮撕取法,比较正常苗与玻璃化苗茎叶横切面显微结构以及叶表面气孔特征。结果表明,黑果腺肋花楸栅栏组织由2~3层细胞构成,下表皮气孔下陷,小而密集,茎内分布大量导管、筛管。玻璃化苗主脉不明显,细胞排列凌乱不规则,栅栏组织与海绵组织的区别不明显,气孔开口大近圆形,茎肿胀粗大髓腔内薄壁细胞较多、已破裂。
Using the tissue culture seedlings of Sorbus aucuparia as test material, the microstructure of stem and leaf of normal seedling and vitrification seedlings and the stomata characteristics of leaf surface were compared by paraffin section and leaf epidermal tear method. The results showed that the tissue of Prunus armeniaca was composed of 2 ~ 3 layers of cells, the stomata of the lower epidermis were subsided, small and dense, and a large number of catheters and screens were distributed in the stem. Vitrification seedling vein is not obvious, irregular arrangement of cells messy, palisade tissue and spongy tissue is not obvious difference, the stomatal opening is nearly circular, thick tuberosity intramedullary parenchyma cells more, has ruptured.
