为观察BTLA mRNA在EAN大鼠坐骨神经、脾脏、外周血及淋巴结中的动态表达,探讨BTLA与EAN的发病关系,以提供治疗自身免疫性疾病新的靶点。采用72只Lewis大鼠,SPF级,雄性,6-8周,体重160~180 g,随机分为生理盐水对照组(NS组)、完全弗氏佐剂组(CFA组)及抗原注射组(EAN组)。观察免疫后大鼠的发病情况和组织病理学改变。分别在第7天、第16天、第24天、第33天处死动物,取坐骨神经根制作病理切片行HE染色和Weil染色观察其病理改变,利用RT-PCR检测坐骨神经、脾脏、外周血和淋巴结中BTLA mRNA的表达水平。结果是EAN组大鼠在发病16d症状达高峰,第33天基本好转。与对照组相比,EAN组大鼠各组织各时间点BTLA mRNA表达均增高(P<0.05),整个实验过程中一直呈上升趋势;CFA组和NS组大鼠无症状,CFA组与NS组相比,BTLA的表达无明显差异。各组织中BTLA的表达具有相关性。提示BTLA在EAN发病过程中的动态变化可能参与EAN的发病。
To observe the dynamic expression of BTLA mRNA in the sciatic nerve, spleen, peripheral blood and lymph nodes in EAN rats and to explore the relationship between BTLA and EAN in order to provide a new target for the treatment of autoimmune diseases. Forty-two Lewis rats, SPF, male, 6-8 weeks old and weighing 160-180 g, were randomly divided into normal saline control group (NS group), complete Freund’s adjuvant group (CFA group) and antigen injection group EAN group). The incidence and histopathological changes of rats after immunization were observed. Animals were killed on the 7th day, the 16th day, the 24th day and the 33rd day respectively. Pathological changes of the sciatic nerve root were made by HE staining and Weil staining. The sciatic nerve, spleen, peripheral blood and lymph nodes were detected by RT- In BTLA mRNA expression levels. The result was that the rats in EAN group reached the peak on the 16th day of symptom onset and basically improved on the 33rd day. Compared with the control group, the expression of BTLA mRNA increased in all groups (P <0.05) at all time points in EAN group and throughout the experiment. The levels of BTLA mRNA in CFA group and NS group were asymptomatic, while those in CFA group and NS group There was no significant difference in the expression of BTLA. The expression of BTLA in each tissue was correlated. It is suggested that the dynamic changes of BTLA in the pathogenesis of EAN may be involved in the pathogenesis of EAN.
