目的:探讨子痫前期患者外周血中IGH-CDR3序列的多样性与免疫学特性。方法:分离子痫前期患者外周血中的B淋巴细胞,逆转录为cDNA后用ARM-PCR扩增H链CDR3区序列,利用高通量测序并与IMGT数据库对比揭示子痫前期患者IGH-CDR3受体库的多样性及免疫学特征。结果:5例子痫前期患者外周血IGH-CDR3的数目和种类差别较大,从1127条read到47385read;子痫前期患者有独有的免疫学特性,包括IGH-CDR3长度、N末端碱基的添加数、V、J区域使用偏好基因及末端去除的碱基数;子痫前期患者外周血中存在某些寡克隆异常增殖的情况。结论:子痫前期患者外周血IGH-CDR3免疫组库有其独有的特点。
Objective: To explore the diversity and immunological characteristics of IGH-CDR3 in peripheral blood of patients with preeclampsia. Methods: B lymphocytes were isolated from peripheral blood of patients with preeclampsia and reverse transcribed into cDNA. The sequence of H chain CDR3 was amplified by ARM-PCR. The sequence of IGH-CDR3 in preeclampsia was detected by high-throughput sequencing and compared with IMGT database Receptor diversity and immunological characteristics. Results: The number and types of IGH-CDR3 in peripheral blood of 5 preeclampsia patients varied greatly from 1127 to 47385read. The patients with preeclampsia had unique immunological characteristics including IGH-CDR3 length, N-terminal base Add the number, V, J regional preference genes and the number of the end of the base removed; peripheral blood of preeclampsia there are some oligoclonal abnormal proliferation. Conclusion: The IGH-CDR3 immune bank of peripheral blood of patients with preeclampsia has its unique characteristics.
