目的:观察高糖对体外培养的大鼠施万细胞半乳糖神经酰胺转移酶(CGT)和半乳糖神经酰胺(GalC)表达的影响。方法:体外培养大鼠施万细胞,用MTT法比较正常葡萄糖(30 mmol·L-1)和高糖(50 mmol·L-1)培养24 h施万细胞的存活力,用半定量RT-PCR 方法测定细胞CGT mDNA表达,用免疫细胞化学方法测定细胞GalC的表达。结果:高糖培养24 h,施万细胞存活力较正常葡萄糖培养时明显下降(73.9%vs 100%,P<0.001);高葡萄糖培养的施万细胞CGT mRNA表达上调:GalC阳性颗粒较正常葡萄糖培养时明显增多。结论:高葡萄糖时施万细胞CGT和GalC表达上调,可能影响施万细胞的活力。
Objective: To observe the effects of high glucose on the expression of galactosylceramide transferase (CGT) and galactosylceramide (GalC) in cultured rat Schwann cells. METHODS: Schwann cells were cultured in vitro. The viability of Schwann cells cultured for 24 h with normal glucose (30 mmol·L-1) and high glucose (50 mmol·L-1) PCR method was used to determine the expression of CGT mDNA, and the expression of GalC was detected by immunocytochemistry. RESULTS: After cultured with high glucose for 24 h, the viability of Schwann cells was significantly decreased compared with that of normal glucose (73.9% vs 100%, P <0.001). The CGT mRNA expression in Schwann cells cultured with high glucose was significantly higher than that of normal glucose Significantly increased when cultured. CONCLUSION: The up-regulation of CGT and GalC expression in Schwann cells during high glucose may affect the viability of Schwann cells.
