目的:观察顺铂肾损伤小鼠后肾组织Caspase-3的表达以及人工冬虫夏草-百令胶囊(Cordyceps sinensis,C.sinensis)干预后对其的影响,探讨C.sinensis的肾保护作用机制。方法:用腹腔注射cisplatin的方法建立顺铂肾损伤模型,C57BL/6小鼠28只随机分为空白对照组、CP组和C.sinensis低剂量、高剂量组,每组7只。72 h后处死小鼠,检测各组小鼠血尿素氮(BUN)和血肌酐(SCr),HE染色观察小鼠肾小管间质病理变化,并用肾小管间质半定量计分法分析各组小鼠病理损伤情况,免疫组织化学检测肾组织Caspase-3的表达。结果:与对照组相比,CP组和C.sinensis低、高剂量组小鼠的BUN及SCr水平增高(P<0.05),肾小管病理半定量计分显示小管间质损伤均明显加重(P<0.05),肾组织Caspase-3表达均明显升高(P<0.05)。与CP组相比较,C.sinensis的低、高剂量组的BUN和SCr水平下降(P<0.05),Caspase-3表达下调(P<0.05),肾小管病理半定量计分显示C.sinensis的低、高剂量组分别较CP组小管间质损伤明显改善(P<0.05)。结论:顺铂肾损伤肾组织中Caspase-3表达明显增高,提示顺铂至急性肾损伤与其诱导肾小管上皮细胞凋亡有关。C.sinensis干预后肾组织中Caspase-3的表达明显降低,提示C.sinensis可能通过抑制肾小管上皮细胞凋亡而达到肾保护作用。
OBJECTIVE: To observe the effect of intervention of Cordyceps sinensis and C. sinensis on the expression of Caspase-3 in the kidney of cisplatin-induced renal injury mice and to explore the renal protective mechanism of C. sinensis. Methods: A cisplatin-induced renal injury model was established by intraperitoneal injection of cisplatin. 28 C57BL / 6 mice were randomly divided into blank control group, CP group and C. sinensis low dose and high dose group, with 7 rats in each group. After 72 h, mice were sacrificed and blood urea nitrogen (BUN) and serum creatinine (SCr) were measured in each group. The pathological changes of tubulointerstitial were observed by HE staining. Semi-quantitative analysis of tubulointerstitial Mice pathological damage, immunohistochemical detection of renal tissue Caspase-3 expression. Results: Compared with the control group, the levels of BUN and SCr in the CP and C.sinensis low and high dose groups were significantly increased (P <0.05), and the semi-quantitative tubular pathological score showed that the tubulointerstitial injury was significantly increased <0.05), while the expression of Caspase-3 in renal tissue was significantly increased (P <0.05). Compared with CP group, the levels of BUN and SCr decreased (P <0.05) and the expression of Caspase-3 decreased (P <0.05) in C.sinensis low and high dose groups. Semiquantitative scoring of renal tubule pathology showed that C. sinensis Compared with CP group, tubulointerstitial injury in low and high dose groups were significantly improved (P <0.05). Conclusion: The expression of Caspase-3 in renal tissue of cisplatin-induced renal injury was significantly increased, suggesting that cisplatin-induced acute renal injury is associated with the apoptosis of renal tubular epithelial cells. The expression of Caspase-3 in kidney tissue decreased significantly after C.sinensis intervention, suggesting that C.sinensis may achieve renal protection by inhibiting renal tubular epithelial cell apoptosis.
