目的:采用正/反向效应评价相结合的研究思路与方法探讨香附四物汤中主要药对的作用特点及对整方的贡献。方法:采用昆明种小鼠作为实验动物,香附-延胡索药对水提取物组、全方除去香附-延胡索药对水提取物组、全方水提取物组、香附-当归药对水提取物组、全方除去香附-当归药对水提取物组按1.17,3.71,5.46,1.75,3.70 g.kg-1剂量ig给药,空白对照ig生理盐水。以小鼠热板法、醋酸致小鼠扭体反应、小鼠原发性痛经模型和小鼠血虚模型,评价各给药组对不同模型小鼠的影响。结果:在3种疼痛模型中,香附-延胡索药对表现出很强的镇痛效果,当其在香附四物汤组方中被剔除后作用明显减弱或消失,而香附-当归药对的镇痛效果相对较弱,当其在香附四物汤组方中被剔除后作用没有明显减弱,有的甚至有所提高。小鼠血虚实验中,各给药组白细胞数、胸脾腺指数与模型组比较无显著性差异,从红细胞数和血红蛋白数指标变化发现,香附-当归药对具有很强的补血效应,而香附四物汤以及当其在香附四物汤组方中被剔除后,其均未表现出明显的补血作用。同时,香附四物汤能非常显著地升高痛经模型小鼠子宫组织中NO的含量,提高SOD活力,并降低Ca2+水平。结论:在香附四物汤中,香附-当归的补血作用减弱,而香附-延胡索的镇痛作用依然显著,并且香附四物汤抑制原发性痛经的作用机制可能与影响子宫组织中NO,Ca2+水平及提高SOD活力有关;同时通过实验发现采用正/反向效应评价相结合的方法是研究方剂配伍效应的有效途径。
OBJECTIVE: To investigate the characteristics of the main drug effects in Xiangfu Siwu Decoction and its contribution to the whole study by using the research ideas and methods of positive / negative effect evaluation. The effects of each administration group on different model mice were evaluated by mouse hot plate method, acetic acid induced writhing reaction in mice, primary dysmenorrhea model in mice and blood deficiency model in mice. Results: In three kinds of pain model, Xiang Fu - Yan Yu drug showed a strong analgesic effect, when it was removed in the Xiang Fu Si Wu Decoction group significantly weakened or disappeared, while the incense - Angelica medicine Right analgesic effect is relatively weak, when it was removed in the side of the package with the four parties did not significantly reduce the role, and some even increased. In mice blood deficiency experiment, there was no significant difference between the white blood cell count and the thymus gland index in each administration group and the model group. According to the red blood cell count and hemoglobin number index, And Fu-Fu Siwu Decoction and when it was removed in Xiangfu Siwu Decoction group, they did not show significant blood effects. Simultaneously, Xiangfu Siwu Decoction can significantly increase NO content in uterine tissue of dysmenorrheal model mice, increase SOD activity and decrease Ca2 + level. Conclusion: In Xiang-Fu-Si-Wu-Tang, the blood-invigorating effect of Xiangfu-Angelica declines, but the analgesic effect of Xiangfu-Fuyanxuo is still significant. The mechanism of Xiangfu Siwu Decoction in inhibiting primary dysmenorrhea may be related to the effect on the uterus tissue In NO and Ca2 + levels and increase SOD activity; at the same time through experiments found that the use of positive / negative effect evaluation of the combination of methods to study the effect of prescription compatibility effect of an effective way.
