目的探讨细粒棘球蚴(Echinococcus granulosus,E.g)感染对小鼠肝脏细胞周期相关蛋白表达的影响。方法C57BL/6J小鼠16只随机分为实验组和对照组,实验组8只以细粒棘球蚴原头节混悬液直视下注射肝脏,对照组8只以相同方法注射无菌PBS,感染180 d后处死小鼠,检测血清中转氨酶的变化,HE染色后观察肝组织病理改变,采用Western blot检测p-ERK、PCNA、cyclin A、cyclin D1和cyclin E在肝内的表达。结果与对照组相比,实验组小鼠血清中ALT和AST分别升高0.34倍和2.33倍;镜下可见肝脏病灶出现坏死、炎性细胞浸润和纤维组织形成等多种病理改变,肝内PCNA蛋白表达增加,且在两组间的表达差异有统计学意义(t=2.37,P<0.05)。结论 E.g感染宿主后,肝脏损伤与感染应激条件下的代偿性增生同时存在。
Objective To investigate the effect of Echinococcus granulosus (E.g) infection on the expression of cell cycle related proteins in mouse liver. Methods Sixteen C57BL / 6J mice were randomly divided into experimental group and control group. In the experimental group, 8 rabbits in the experimental group were directly injected with the suspension of Echinococcus granulosus and the control group were injected with sterile PBS The mice were killed after 180 days of infection and the changes of serum aminotransferase were detected. The pathological changes of liver were observed by HE staining. The expressions of p-ERK, PCNA, cyclin A, cyclin D1 and cyclin E in the liver were detected by Western blot. Results Compared with the control group, the serum ALT and AST in experimental group increased by 0.34 times and 2.33 times, respectively. The pathological changes such as necrosis, inflammatory cell infiltration and fibrous tissue formation were found in the liver lesions. The protein expression increased, and the difference between the two groups was statistically significant (t = 2.37, P <0.05). Conclusion After E.g host infection, hepatic injury coexisted with compensatory hyperplasia under infection stress.
