目的 探讨人类蔗糖非发酵蛋白2同源物(human sucrose nonfermenting protein 2 homologue,hSNF2H)在非小细胞肺癌(NSCLC,non-small cell lung cancer)中的表达情况,分析其与临床病理学特征的关系.方法 采用免疫组化SP法检测88例NSCLC中hSNF2H的表达和定位,Western Blot检测hSNF2H在正常支气管上皮细胞HBE和肺癌细胞A549、H1299和LET中的表达情况,Transwell的方法检测干扰hSNF2H的表达对肺癌细胞A549和H1299侵袭和转移能力的影响.结果 hSNF2H在NSCLC表达的阳性率(59.1％,52/88)显著高于癌旁肺组织(5.7％,5/88,P＜0.001);hSNF2H在肺癌细胞中的表达水平显著高于HBE细胞(P＜0.001),与患者的TNM分期(P=0.014)和淋巴结转移(P=0.008)密切相关.干扰hSNF2H可显著抑制A549和H1299细胞的侵袭和迁移能力.结论 hSNF2H参与非小细胞肺癌的发病机制.“,”Objective To explore the expression of human sucrose nonfermenting protein 2 homologue (hSNF2H) in numan non-small cell lung cancer (NSCLC) and its correlation with clinicopathological characteristics.Methods SP (streptavidin-perosidase) immunohistochemistry was used to detect the expression of hSNF2H in 88 cases of NSCLC and their adjacent noncancerous lung tissues.Western blot was used to detect the expression of hSNF2H in HBE(normal bronchial epithelial cell line) and three lung cancer cell lines A549,H1299 and LTE,and the effects of hSNF2H interference on invasion and migration abilities of lung cancer cells detected by transwell.Results The positive expression ratio of hSNF2H in lung cancer tissues was 59.1％ (52/88),significantly higher than that in adjacent lung tissue(5.7％,5/88,P＜0.001);the expression level of hSNF2H in three lung cancer cells was significantly higher than that in HBE (P＜0.01),related to TNM stage (P=0.014) and lymph node metastasis(P=0.008) of lung cancer patients;and interfering with hSNF2H could significantly inhibit the invasion and migration abilities of A549 and H1299 cell lines.Conclusion hSNF2H might be involved in the pathogenesis of NSCLC.
