[目的]研究SIAH2和HIF-1α蛋白在乳腺浸润性导管癌组织中的表达及其与临床病理特征的关系。[方法]采用免疫组织化学方法检测80例乳腺浸润性导管癌组织和10例乳腺癌旁正常组织中SIAH2和HIF-1α的表达;采用Western Blot法检测25例乳腺浸润性导管癌和20例乳腺纤维腺瘤中SIAH2和HIF-1α蛋白的表达。[结果]乳腺浸润性导管癌组织中SIAH2及HIF-1α的阳性表达率分别为60%和65%,癌旁正常组织中均未见表达。SIAH2和HIF-1α表达存在正相关关系(r=0.310,P<0.05)。两种蛋白表达与肿瘤的组织学分级、临床分期和淋巴结转移密切相关,而与肿瘤大小、年龄无显著相关性。Western Blot结果表明SIAH2和HIF-1α两种蛋白在乳腺癌组织中的表达均高于乳腺纤维腺瘤中的表达(P均为0.00)。[结论]SIAH2及HIF-1α过表达在乳腺癌的发展及浸润转移中具有重要的作用。
[Objective] To investigate the expression of SIAH2 and HIF-1α in breast invasive ductal carcinoma and its relationship with clinicopathological features. [Method] Immunohistochemical method was used to detect the expression of SIAH2 and HIF-1α in 80 cases of breast invasive ductal carcinoma tissues and 10 cases of normal breast tissues. The expression of SIAH2 and HIF-1α in 25 cases of breast invasive ductal carcinoma and 20 cases of breast carcinoma Expression of SIAH2 and HIF-1α in Fibroadenoma. [Results] The positive expression rates of SIAH2 and HIF-1α in invasive ductal carcinoma of the breast were 60% and 65% respectively, and no expression was found in the normal tissues adjacent to the tumor. There was a positive correlation between SIAH2 and HIF-1α expression (r = 0.310, P <0.05). The expression of the two proteins was closely related to histological grade, clinical stage and lymph node metastasis, but not to tumor size and age. The results of Western Blot showed that the expression of both SIAH2 and HIF-1α in breast cancer was higher than that in breast fibroadenoma (P = 0.00). [Conclusion] SIAH2 and HIF-1α overexpression play an important role in the development of breast cancer and invasion and metastasis.
