目的建立HPLC同时测定功能红曲中红曲可林K(monacolin K)、红曲可林L(monacolin L)和脱水红曲可林K(dehydromonacolin K)的方法。方法色谱柱为Agilent Zorbax SB C18柱(250 mm×4.6 mm,5μm);流动相为甲醇-0.05%磷酸(78∶22);检测波长238 nm;体积流量1.0 mL/min;柱温35℃;进样量20μL。结果红曲可林K、红曲可林L和脱水红曲可林K的线性范围分别为0.08~0.84、0.04~0.38、0.04~0.40μg;加样回收率分别为99.2%、99.6%、99.3%,RSD分别为1.7%、1.5%、1.4%(n=9)。结论本方法简便、灵活、准确,重复性好,可用于功能红曲中红曲可林K、红曲可林L和脱水红曲可林K的测定。
Objective To establish a HPLC method for simultaneous determination of monacolin K,monacolin L and dehydromonacolin K in red yeast rice. Methods The chromatographic column was Agilent Zorbax SB C18 column (250 mm×4.6 mm, 5 μm); the mobile phase was methanol-0.05% phosphoric acid (78∶22); the detection wavelength was 238 nm; the volumetric flow rate was 1.0 mL/min; the column temperature was 35°C; Injection volume 20μL. Results The linear ranges of Hongqu Kelin K, Hongqu Kelin L and dehydrated Red Kobolin K were 0.08~0.84, 0.04~0.38 and 0.04~0.40μg, respectively. The recoveries of the samples were 99.2%, 99.6% and 99.3 respectively. %, RSD 1.7%, 1.5%, 1.4% (n=9). Conclusion The method is simple, flexible, accurate, and reproducible. It can be used for the determination of Monascin K, Mongoline L, and Monascus K in functional red yeast rice.
