目的观察2型糖尿病大鼠肌肉组织中过氧化物酶体增殖物激活受体2(PPAR-γ2)的表达,探讨2型糖尿病的病变机制。方法 SD大鼠随机分为正常对照组和模型组,各10只。模型组高脂饲料喂养2个月后,腹腔注射链脲佐菌素(STZ,40 mg.kg-1)复制糖尿病模型,采用RT-PCR方法,检测造模后6周肌肉组织PPAR-γ2 mRNA的表达。结果造模后6周,糖尿病模型组大鼠肌肉组织PPAR-γ2 mRNA的表达明显低于正常对照组(P<0.01)。结论 2型糖尿病伴有大鼠肌肉组织PPAR-γ2的表达水平下降。
Objective To observe the expression of peroxisome proliferator - activated receptor 2 (PPAR - γ2) in the muscle tissue of type 2 diabetic rats and explore the mechanism of the pathogenesis of type 2 diabetes mellitus. Methods SD rats were randomly divided into normal control group and model group, with 10 rats each. At 2 months after model group fed with high-fat diet, diabetic model was induced by intraperitoneal injection of streptozotocin (STZ, 40 mg.kg-1), and RT-PCR was used to detect the expression of PPAR-γ2 mRNA expression. Results Six weeks after model establishment, the expression of PPAR-γ2 mRNA in muscle tissue of diabetic model group was significantly lower than that of the normal control group (P <0.01). Conclusions The expression of PPAR-γ2 in rats with type 2 diabetes mellitus is decreased.
