本研究以a—~(32)P—dCTP标记的HPV—16基因组中三组亚基因片段为探针,应用制(?)内切酶技术和Southern印迹杂交方法进一步分析了9例HPV—16基因组已发生整合宫颈癌组织DNA中HPV—16基因组各亚基因片段存在的情况。结果表明,在整合状态下,PV—1基因组的部分L1 L2区和大部分E2 E4区是缺失的,而HPV—16基因组的E6 E7区■完整地被保留下来。提示HPV—16基因组的缺失区域很可能是其基因组发生断裂整合部位,而HPV—16基因组的E6 E7区可能是其导致细胞恶性转化的致癌基因。 In this study, three subgroups of HPV-16 genomes labeled with a- ~ (32) P-dCTP were used as probes. Nine HPV-16 gene fragments were further analyzed by using restriction endonuclease digestion and Southern blotting. Genomic integration of cervical cancer tissue has occurred in the DNA HPV-16 genome sub-gene fragments exist. The results showed that part of the L1 L2 region and most of the E2 E4 region of the PV-1 genome were deleted while the E6 E7 region of the HPV-16 genome was completely preserved in the integrated state. It is very likely that the deletion region of HPV-16 genome is likely to have a cleavage site in its genome. However, the E6 E7 region of HPV-16 genome may be the oncogene that leads to the malignant transformation of HPV-16 genome.
